Kindly asking to clarify if its possible to change Traffic script rule via API request (vTM 21.3).

From the vTM 21.3: Control API Guide (p.395)

The Catalog.Rule interface in the API allows management of TrafficScript Rules.  Using this interface, you can create, delete and rename rules, and manage their configuration.  You can also syntax-check rule fragments.

In addition, the Traffic Manager REST API Configuration resource is fully interactive and maps directly to the native Traffic Manager configuration system.  Each concept, such as pools, virtual servers, TrafficScript rules, or Service Level Monitoring classes, has an associated configuration resource model.
